Kumtorn Nateethanasarn, a coordinator of the event, said the run was neither for nor against the government's plans to build a dam in the park.
He said it was organised to allow the participants to see for themselves if a dam should be built in the area.
He said most people received information about the dam project from media outlets. Local people seemed divided over its merits.
Mr Kumtorn said the run kicked off at 11pm on Friday at Lumpini Park and ended Sunday at Mae Rewa Wildlife Sanctuary which is part of the national park. The event was joined by about 200 people.
