Goals from Suchao Nutnum after 39 minutes and Charyl Chappuis finished off the Chinese visitors at I-Mobile Stadium.
In the other Group E match, Japan's Vegalta Sendai defeated South Korea's FC Seoul 1-0 with Atsushi Yanagisawa scoring the winner in the 16th minute.
The Thai FA Cup champions moved from bottom of Group E to joint second with Sendai on five points. FC Seoul lead the pool with seven points, while Jiangsu have four.
Buriram, who lost 2-0 at Jiangsu last week, started strongly and attacked Jiangsu from the opening whistle.
After squandering several chances, Buriram broke the deadlock in the 39th minute when poor defending allowed an unmarked Suchao to head the ball home past goalkeeper Deng Xiaofei.
The Thunder Castle were still the dominant side in the second half and deservedly got their second goal in the 59th minute when Thai-Swiss Chappuis received a pass from Japanese Kai Hirano to score.
Unfortunately, Chappuis was injured while celebrating and was replaced by Thai-American Anthony Ampaipitakwong.
Jiangsu regrouped and was better, but could not break Buriram's defence from two goals down.
